Test Method:

1. Instruments and reagents: LC-10AVP high performance liquid chromatograph; SPD-10AVP detector; Acetonitrile (chromatographic pure)
2. Operation steps: according to high performance liquid chromatography determination.
a. Chromatographic conditions: octadecylsilane bonded silica gel was used as filler (150*4.6mm*5.0μm); Using Acetonitrile: Water = 3:2 as mobile phase; The detection wavelength was 258nm. Column temperature: 25℃; Flow rate: 0.5ml/min; The number of theoretical plates should be no less than 2000 according to the main peak of PyBOP.
b. Determination method: Take this product and add mobile phase to make a solution containing PyBOP 100ug per 1ml as the test product solution. 20μl of the test solution was injected into the chromatograph, and the chromatogram was recorded to 2 times the retention time of the principal component peak.
c. Results: This product is calculated as a dry product, containing no less than 99.0% PyBOP.

Application:

1H-Benzotriazol-1-yloxytripyrrolidinophosphonium Hexafluorophosphate (PyBOP) (CAS: 128625-52-5)is one of the most convenient peptide coupling reagents utilized in solid phase peptide synthesis without carcinogenic by-products. It is also used as a substitute for the (benzotriazol-1-yloxy)tris(dimethylamino)phosphonium hexafluorophosphate (BOP) reagent. It has reactivity similar to BOP, but its byproduct is not highly carcinogenic. PyBOP is especially suitable for solid-phase peptide synthesis, avoids racemization. In contrast to uronium-based coupling reagents, PyBOP does not form guanidinated byproducts when used in excess. This makes it useful in fragment condensation reactions and head-to-tail cyclizations.
Analogs of BOP coupling reagents, but do not form carcinogens HMPA is a by-product. May be used in place of BOP in peptide synthesis without loss of coupling efficiency. Coupling reactions are rapid, being nearly complete within a few minutes. Especially useful for cyclization steps for the activation of hindered amino acid.
