Tadalafil was marketed in the United States in December 2003 as a daily oral dosage form. It is also approved in the United States and Europe for benign prostatic hyperplasia and pulmonary hypertension.

Preparation of D-Tryptophan Methyl Ester Hydrochloride: D-Tryptophan (50g) is added to the mixed solution of toluene (650ml) and methanol (105ml) and stirred evenly, slowly dropwise addition of thionyl chloride (38.6g) at 0 degrees Celsius, after the dropwise addition, the temperature is raised to 75-80 degrees Celsius for 4 hours, the reaction is over, the reaction liquid is cooled to 0 degrees, the solid was precipitated, filtered, the filter cake was washed with toluene, and dried in vacuum at 50℃ for 6h to obtain D-Tryptophan Methyl Ester Hydrochloride. The yield was 95% and the HPLC purity was 99%.
