This standard operating procedure is intended to set down the correct method of analysing the purity of a compound that contains a basic amino group. eg. Benzylamine.
Health and safety
When sampling chemicals min PPE: lab coat, safety specs and gloves should be worn
It is not acceptable to wear less than the minimum required level of PPE.
Ensure that the fume cupboards are working properly – if in doubt consult supervisor/manager.
Running the Titration
Titrations must always be done in duplicate.
Clearly mark 2 x 250ml conical flasks with numbers 1 and 2
Ensure flasks are clean.
Add 120ml of acetic acid to one clean dry flask and 2 ml of Naptholbenzein indictor (pre made in acetic acid)
Neutralise the acetic acid using 0.1M Perchloric acid until the colour goes from yellow to deep green. (1 drop end point)
Divide the neutralised acetic acid equally between the flasks and ensure the necks of each flask are free from liquid.
Dissolve about 2mmol equivalents of your sample, weighed accurately to 4 decimal places into one of your pre neutralised conical flasks.
Record your weighing.
Ensure that the material fully dissolves in the acetic acid, the solution should go from deep green to bright yellow.
If your sample does not dissolve in acetic acid you may need to use a small amount of formic acid to dissolve the sample before adding the acetic acid.
Titrate each sample against 0.1M Perchloric acid, until the deep green end point colour is reached.
Record the volume of 0.1M Perchloric acid used
Repeat this process with your second sample
Calculation of the purity
Assay of product = V × c x (FW/1000)÷ W × 100
where V = Volume (ml) of 0.1M perchloric acid
W = Weight of sample (g)
c = Concentration of perchloric acid(in this case 0.1)
Duplicated results must be within 0.5% of one another.
If the results you receive are not within 0.5% then the procedure must be repeated until you achieve this.
