Application:

4-(Acetoxymethyl)benzoic Acid (Ac-HMBA-Linker) (CAS: 15561-46-3), linkers for solid phase synthesis. A base labile 4-hydroxymethylbenzoic acid (HMBA) linker,it was effectively removed at pH=10. The HMBA linker was demonstrated to be stable to the conditions required for native chemical ligation, subsequent transformations and final purification. The benzyl ester, which forms the HMBA linker with the first amino acid, is stable to piperidine, used for Fmoc group removal, making the linkage applicable for chemical elongation of a peptide sequence with Fmoc strategies. A large variety of C-terminally modified peptides was obtained by nucleophilic cleavage of the ester bond in solid phase linked peptide esters of 4-hydroxymethyl benzamide (HMBA). The developed methods provided peptides, C-terminally functionalized as esters, amides and thioesters, with high purity directly from the resin in a single reaction step. 4-hydroxymethylbenzoic acid linkage, which was cleaved by ammonolysis in isopropanol, gave the model peptide 1 in good overall yield (53% including functionalization).
